GENERAL REMARKS:
Somatotropin is the major growth hormone, produced in the anterior lobe of pituitary glands in mammals, and in humans is approximately 10% of the dry weight of the gland. The human growth hormone is sometimes given the more specific name "somatropin", with the CAS number 12629-01-5(1,2,4), although the growth hormone may also be referenced by species-specific or (for recombinant forms) source-specific CAS numbers.
An untreated deficiency of the hormone produces dwarfism. Excessive hormone produced before puberty results in gigantism; if excessive hormone is produced after puberty, the resulting condition is called acromegaly. Treatment with HGH (natural or recombinant) or an analog "somatrem" can be effective; somatrem differs by an additional methionine residue5 and may be more antigenic.(1)

STRUCTURE, PHYSICAL PROPERTIES:
The structure of human somatotropin is reported as either 190(3) or 191(1,2,4) amino acid residues with two disulfide bridges (at 53/165 and 182/189), with molecular weight of approximately 22 kDa. The isoelectric point (pI) has been reported as 4.9.(2,3) Neither pI nor molecular weight have been verified by Sigma. The reported sequences for somatotropin and somatrem are provided for comparison on the end of this document.

DESCRIPTION OF THE PRODUCT AS SOLD:
Each vial of S4776 contains 2 mg somatotropin, 0.1 mg ammonium hydrogen carbonate and 3.5 mg mannitol, lyophilized from a solution at pH 7.95. (Product S1774 from the same supplier was offered in a different buffer.) Contamination by other glycoproteins is low, but is no longer reported for each lot. (Typical values ​​for S1774, reported as maximum: h-luteinizing hormone, 1%; h-thyroid stimulating hormone, 1%; h-follicle stimulating hormone, 1%; h-prolactin, 2%; h-ACTH, 1%). (3)
This growth hormone was standardized against a World Health Organization Standard (stated on certificate of analysis). Its activity was measured by radioimmunoassay using an antiserum raised in a rabbit by immunization with eliminated human growth hormone. STABILITY:
Somatotropin should be stable in the lyophilized form for at least a year.

SOLUTIONS:
The contents of a vial should be dissolved with 1 mL of deionized water by gentle mixing (swirling). This solution can then be stored between 2-8?C for 3-4 weeks. Solutions should not be frozen and thawed.
